Diabetes mellitus have become the third leading chronic disease among the population, following cancer and cardiovascular disease (CVD). Overview of T2DM

Type 2 diabetes mellitus (T2DM) patients account for about 90% of all diabetic patients. Over 2.2 billion people were overweight, with over 774 million of those being obese. Type 2 diabetes, which is often exacerbated by obesity, was estimated to afflict 463 million people worldwide. Developing therapies to combat these metabolic diseases is in urgent need, as is the need for animal models to test the efficacy of such therapies.

Model Description
Ob/ob Mouse The model is a monogenic model of T2DM. The animals are homozygous for a mutation in leptin and develop obesity, hyperphagia, glucose intolerance, insulin resistance and transient hyperglycemia.
Db/db Mouse The model is also monogenic, insulin-resistant model of type 2 diabetes, due to a spontaneous mutation in the leptin receptor. Similar to the ob/ob strain, the animals are obese, hyperinsulinemia (for a portion of their lives), insulin resistant, and hyperglycemic.
KK-Ay Mouse KK-Ay is another diabetes model whose obesity is driven by hyperphagia. The mice, however, do not become as morbidly obese as ob/ob and db/db mice. KK-Ay mice develop impaired glucose tolerance and insulin resistance, and fasting hyperglycemia.
Diet-induced Obesity (DIO) Mouse Among mouse strains, C57BL/6 is among the most sensitive to this diet-induced obesity (DIO). C57BL/6-DIO mice, particularly males, develop severe obesity, increased glucose intolerance, moderate insulin resistance and elevated glucose levels.
NZO Mouse NZO mouse is a polygenic model of obesity, created by selective breeding.
TallyHo Mouse It is a naturally occurring model of obesity and T2DM derived from selective breeding of mice.

NOD mouse models are an inbred, genetically well-characterized mouse strain. Female mice have a propensity to develop T1DM via an autoimmune pathway, making them particularly useful for dissecting the immunologic aspects and the role of immune tolerance in the development of T1DM.
