Gestational diabetes mellitus (GDM) is a common mid- to late-term pregnancy disorder characterized by hyperglycemia, hyperinsulinemia, and impaired glucose tolerance. Gestational diabetes is thought to be multifactorial, stemming from genetic and environmental factors. High-saturated-fat diet gestational diabetes mellitus mouse models are the most widely used mouse models for studying gestational diabetes, which closely simulates metabolic abnormalities seen in human GDM.

The HFD-GDM mouse is basically generated by feeding female mouse with HFD (60% fat) for few weeks, then mating them with male mouse, and the embryonic day 0.5 was designated once the vaginal plug was present. The recommend control of HFD-GDM mouse is normal diet (10% fat) mouse.
