Imaging of Microglia Activation in Animal Models of Stroke

Microglia are the resident macrophages of the central nervous system. They play critical roles in phagocytosis of cellular debris, presentation of foreign antigens, and as key sensors of pathological events such as ischemia. Activation of microglia after ischemic stroke involves several stereotypical events, including morphological transformation, proliferation, and polarization. However, the precise functional roles of activated microglia in ischemic stroke have not been fully elucidated. Studies have shown that activated microglia display different polarization phenotypes and that different events may occur during the complicated process of microglial activation under different ischemic conditions.

Fig. 1. The response of microglial processes to laser-induced damage demonstrates a time-dependent specificity, particularly within the thalamus.Fig. 1 Microglia process response to laser damage over time shows a specificity to the thalamus. (Kluge, et al., 2019)

Our Microglial Dynamics Assessment Services

Ace Therapeutics provides comprehensive microglial dynamics assessment services to help clients assess the morphology of microglia, their process motility, and migration in animal models of stroke. Our microglial dynamics assessment services are designed to meet the needs of both basic research and drug screening, as well as efficacy evaluation. We provide comprehensive technical support to facilitate the development of innovative stroke therapies.

We use two-photon microscopy technology to perform in vivo brain imaging in live animals, providing high-resolution visualization of microglia with the following readouts:

  • Microglia morphology
  • Microglia process motility
  • Microglial migration
  • Colocalization of BBB-penetrating antibodies with microglia and sub-cellular organelles in microglia

Advantages of Our Microglial Dynamics Assessment Services

  • In vivo imaging: We can image the brain of live animals after a stroke in depth and at high resolution.
  • Comprehensive readouts: In addition to microglial dynamics assessment, we offer the flexibility to incorporate other critical readouts, such as behavioral assessments, drug cross BBB, and BBB integrity.
  • Longitudinal monitoring: Our advanced imaging techniques allow us to assess microglial dynamics over hours, days, weeks, or even months after stroke.

Our Microglial Dynamics Assessment Services Are Applicable To

  • Study the roles of microglia in neuroinflammation, neuroprotection, and neural repair mechanisms in stroke.
  • Evaluate the effects of potential drugs on microglial activity and function to identify drug candidates with neuroprotective properties.

Leveraging advanced imaging technologies and extensive expertise in stroke research, Ace Therapeutics has launched microglial dynamics assessment services. We aim to provide clients with high-resolution, real-time observation of microglial dynamics to facilitate in-depth studies of stroke pathology and the development of novel therapeutic strategies.
