3D BBB Models Culture Substrate

3D BBB Models Culture Substrate

The BBB is a specialized barrier in the brain that regulates the entry of molecules and cells from the bloodstream into the brain. It plays a crucial role in protecting the brain from harmful substances and maintaining its homeostasis.

Traditional 2D cell model of BBB culture is performed on simple, nonporous two-dimensional (2D) surfaces. Despite the method being easy and convenient, can result in certain well-documented artifacts. The 3D BBB model culture substrate refers to the three-dimensional culture system used for culturing and studying the blood-brain barrier (BBB) model in vitro, which aims to mimic the physiological conditions of the brain's microenvironment, allowing researchers to study BBB function and dysfunction in a controlled laboratory setting. It typically consists of a specialized culture substrate that supports the growth and differentiation of brain endothelial cells, which form the main component of the BBB. The substrate is often coated with extracellular matrix proteins like collagen or laminin to provide a suitable surface for cell attachment and growth.

A well-established method for culturing 3D BBB models is to culture cells on structural scaffolds, which are typically composed of biopolymers, such as 3D hydrogels and collagens, arranged to mimic the physiological extracellular matrix (ECM).
