Gene Description Metaxin-1 and Metaxin-2 are part of the sorting and assembly machinery that has been associated with import of β-barrel proteins through the mitochondrial outer membrane. MTX1 and MTX2 can also function in mitochondrial trafficking in neurons. Studies of cells with a loss of MTX1 have shown that MTX1 is required for some forms of cell death, including TNF-α-induced cell death. Subsequent studies found that MTX1 and MTX2 can regulate TNF-α mediated apoptosis through association with the apoptotic effector protein Bak, and that phosphorylation of MTX1 controls Bak activation. MTX1 has also been shown to be targeted for basal piecemeal mitophagy in an LC3C- and p62/SQSMT1-dependent manner. MTX1 was identified as a regulator of sorafenib resistance in hepatocellular carcinoma by augmenting autophagy.
