Gene Description Hepcidin is a secreted peptide hormone that functions as a master regulator of systemic iron homeostasis. Hepcidin is synthesized as a preprohormone, primarily by hepatocytes; the preprohormone undergoes a series of cleavage events, culminating in the release of mature hepcidin into circulation, where it plays a critical role in regulating plasma iron levels. Hepcidin in circulation binds to ferroportin, a channel protein that regulates cellular iron export, and whose expression levels are post-translationally regulated via its interactions with hepcidin. Chronic overexpression of hepcidin leads to iron-restricted anemia in mammals, whereas hepcidin deficiency leads to disease states associated with iron overload. Hepcidin levels can thus serve as useful biomarkers for various disease states related to iron homeostasis.
