Gene Description Changes in mitochondrial dynamics regulated by environmental cues affect mitochondrial size and shape and have been shown to dramatically impact mitochondrial metabolism, apoptosis, and autophagy. Mitochondrial fusion and fission are dynamically controlled by several interacting proteins. Mitochondrial fission is controlled by the GTPase DRP1 which is recruited to the mitochondria by FIS1 and MFF. FIS1 is localized to the mitochondrial outer membrane and overexpression induces fission and leads to apoptosis. Decreased expression of FIS1 leads to mitochondrial elongation and triggers cellular senescence. Elevated expression of FIS1 can also promote autophagy. Studies also found that FIS1 interacts with the autophagy initiation protein STX17 and that depletion of FIS1 leads to an increase in mitochondrial localized STX17 and mitophagy. Expression of FIS1 has been associated with neurodegeneration and disease, including cancer.
